Glucocorticoids are widely used as neuroprotective drugs for spinal cord injury. However, the mechanism of neuroprotective effect of glucocorticoids has not been fully elucidated. In the present study, we examined the effects of methylprednisolone (MP) on the generation and maturation of neurons in the subventricular zone (SVZ) in rats subjected to spinal cord injury (SCI). SCI was induced by an injection of 25 microL of 0.9% NaCl into the T-10 segment, and the rats were randomly divided into four groups: sham-operated rats and rats treated with saline, MP, or MP plus megesterol acetate (MPA). MP or MPA was administered by intraperitoneal injection of 200 mg/kg once daily from 3 days to 8 weeks. There were three control groups: (1) the spinal cord-injured-saline (SCIS) group, (2) the SCIS plus MP group, and (3) the SCIS plus MPA group. The therapeutic effect was evaluated by locomotor function, thermal sensitivity, and Basso-Beattie-Bresnahan (BBB) rating scale tests at 1, 2, 3, 4, 5, 6, 7, 8, and 9 weeks after SCI. Spinal cord tissue was stained with cresyl violet, and sections were photographed using a microscope. Vimentin-immunoreactive and nestin-immunoreactive cells, B-cell lymphoma 2 (Bcl-2), and Bcl-2-associated X (Bax) proteins were identified by immunohistochemistry. The migrating and migrating distance of the newly formed cells were counted by Image J, and the percentage of nestin-immunoreactive cells, vimentin-immunoreactive cells, Bcl-2, and Bax were determined. The results showed that the number of nestin-immunoreactive cells and the percentage of nestin-immunoreactive cells, both of which were significantly increased in the MP and MPA treatment groups (P < 0.05), were markedly higher than those in the SCIS and SCIS plus MP groups at 2, 3, 4, 5, 6, 7, 8, and 9 weeks after
