Lupines are common legumes in China. In mid-May of 2009, a lupine (Lupinus rufillosus) field was found in Jinggangshan (Jiangxi Province, China), exhibiting yellow streaks in leaves. Thirty other plants in the field exhibited similar symptoms. Streak symptoms were characterized by a yellowing of the lamina followed by a necrosis and chlorosis of the affected area. The streaks began at the leaf edges and then expanded to the midrib with the lower leaves showing the symptoms more severely. In about 1 month, the leaf blades became shriveled and the plants eventually died. Petioles and stems were also affected. A consistent bacterial growth was observed in streak symptoms after streaking a 1-cm diameter cephalothin-containing disk onto R2A medium. Single colonies grown on R2A medium were purified twice by streaking three times on the same medium. Bacterial morphology, including size and shape, was examined by Gram-staining and light microscopy (LM). Bacterial colonies produced bright fluorescent pigments, which were more prominent under epifluorescence (EF) and seemed to fluoresce blue on LM. Colonies were nonfluorescent when stained with the methylene blue (MB) fluorescent stain. The cells were rod-shaped (0.6 to 1.2 μm in width and 1.1 to 2.4 μm in length) and motile with either single or multiple flagella. Biochemical identification based on catalase (CAT), oxidase (OX), and DNase activities was performed. Total genomic DNA was isolated using a cetyltrimethylammonium bromide-based DNA extraction method (Sigma, St. Louis, MO) and used as the template.
