Murine monoclonal antibodies (MAbs) were generated against the human breast cancer-associated glycoprotein, HER2/neu, a 185-kDa tyrosine kinase-associated transmembrane glycoprotein whose amplification and overexpression have been linked to tumorigenesis. Immunohistochemistry (IHC) of a variety of murine and human mammary tumors, as well as of various carcinomas of the breast and other organs, with the MAbs reveals exclusive (97%) reactivity with human HER2/neu protein. The MAbs were generated by the fusion of NS-1 myeloma cells with spleen cells from BALB/c mice immunized with human HER2/neu-overexpressing BT-474 human breast carcinoma cells. Western blotting of cell lysates with the anti-HER2/neu MAbs distinguishes the 185-kDa protein from its close homologue HER1/EGFR. The recognition of the 185-kDa band by the MAbs was blocked by preincubation of the MAbs with the recombinant, proteolytically cleaved form of the protein (p185HER2/neu), as well as with the recombinant p185HER2/neu-derived extracellular domain. No reactivity was detected with the recombinant transmembrane-cytoplasmic domain of p185HER2/neu. The MAbs stained BT-474 cells at the native surface density, but also internalized and accumulated in the cytoplasm and nuclei of the HER2/neu overexpressing cell. This pattern of distribution of the MAbs was observed in various other human carcinoma cell lines and, thus, the MAbs may prove useful for studying the role of p185HER2/neu in growth regulation, invasion, and metastasis of cancers.
